This study uses hyperthermophilic hydrolysis (HTH) as a pretreatment to boost methane-rich biogas yields from recalcitrant agricultural residues. The hyperthermophilic anaerobe Caldicellulosiruptor bescii hydrolyzes lignocellulose into hydrogen, simple sugars, and organic acids, accelerating downstream anaerobic digestion. Using dairy manure and wheat straw, the project evaluates operational variables and feedstock characteristics to optimize the balance between hydrogen production and methane yield, advancing valorization of an underused waste stream as renewable energy.

📄 Abstract(原文)
Traditional anaerobic digestion (AD) of agricultural waste such as crop residues results in low biogas quality and yield due to their recalcitrant nature, often requiring extended retention times. To increase methane rich biogas yield from crop residues, hyperthermophilic hydrolysis (HTH) can be used a pretreatment step prior to AD. HTH enhances the biodegradability of agricultural waste and demonstrates a circular solution for conversion of waste into clean bioenergy. Hydrolysis is often a rate limiting step in AD for lignocellulosic substrates such as crop residues due to their complex carbohydrate structure. This study will evaluate a hyperthermophilic obligate anaerobe called Caldicellulosiruptor bescii that is capable of enzymatically hydrolyzing lignocellulosic biomass, subsequently producing hydrogen, simple sugars, and organic acids. HTH will leverage microbes specifically adapted to survive and grow at high temperatures to break down the cellulose and hemicellulose present in crop residues, into simple sugars than can quickly be converted into methane during AD. This methane rich biogas, produced at a rapid rate, can then be combusted to produce heat or electricity. Additionally, utilizing an HTH pretreatment process with C. bescii allows for the production and capture of hydrogen, that would otherwise be consumed by methanogens in traditional AD. Hydrogen is a valuable energy product on its own, can enhance methane yield when fed into an AD, or be supplied to a generator for electricity generation. Determining the optimal balance of hydrogen production and downstream methane yields to increase the economic viability of the HTH process, will support its use as an effective method for the overall conversion of agricultural waste to energy. This project will investigate HTH combined with AD to convert dairy cattle manure and wheat straw into methane by evaluating key operational variables, and feedstock characteristics that could influence HTH's performance. The research aims to substantially advance the valorization of an under-leveraged waste stream for the production of methane rich biogas which can be used as a renewable energy source to displace fossil fuel consumption.
